Delivery, installation and training incl. warranty of an Automated Multiplex Imaging Plattform
Technical Must-Criteria which must be met by the System:
Must be a fluorescence-based automated high-content staining and imaging platform for deep phenotyping and spatial biology
System must enable fully-automated and sequential ultrahigh-content image analysis (>500 marker) and allow staining on at least 4 independent tissue slides per run
System must be a fully automated complete platform including instrument, sample carriers, analysis software, system reagents and prevalidated optimized antibody reagent panels and suitable to be offered for service in a core facility
System must allow for technically unlimited cycles of staining, imaging, bleaching and technically unlimited number of parameters
System must allow the staining of FFPE and fresh frozen tissue sections as well as adherent and non-adherent cell preparations
Technical MUST Criteria continued:
An extensive catalogue of manufacturer-prevalidated antibodies (>1000) must be available to work on FFPE and fresh-frozen sections or cells fixed with PFA or acetone
User must however also be free to choose suitable directly-conjugated antibodies of any species (DAPI/FITC/PE/APC or similar) from any vendor on the market to build own panels or add to pre-existing validated reagents (open source reagents)
Sample processing and imaging must not damage the sample/section but allow for further downstream experiments on the same sample/section
Future use as a multiomics-analyzer for simultaneous measurement of protein and RNA must be possible without a change in instrument hardware
Use as a multiomics analyzer must be achieved by a software-only upgrade which is to be included free of charge in the package for at least 3 years
Must be delivered with a matching instrument table
Must be equipped with a Widefield epifluorescence microscope
Must be equipped with at least 3 objectives for overview and working distance images: 2×, 20× long working distance, 20× objective with high numerical aperture
Must have an imaging area of 17x47 mm or bigger
Must be equipped with a dual autofocus (hardware and software based) to ensure consistently sharp imagery also on uneven sample material
Must be equipped with a sensitive sCMOS camera
Must enable analysis at single cell level with a subcellular resolution of <600 nm or below.
Must enable excitation of commonly used fluorochromes in the range of 375 to to 740 nm
Must be equipped with at least five non-overlapping emission filters for commonly used fluorochromes in a range from 450 nm to 840 nm.
Must contain a separate photobleaching unit in combination with fully automatic enzymatic bleaching option
Must be equipped with an automated sample stage with sub-micron positioning accuracy of 100 nm or better
Must allow the use of sample carriers of various sizes (such as microscopy slides, 24/96-well plates or deep well plates) to enable users to measure thin tissue sections as well as adherent tissue culture cells
Must have two computers (with 25TB storage) and screens
Must include instrument operational software and two independent permanent academic software licenses
Must allow high dimensional data analysis of several hundered parameters, k-means clustering, UMAP and t-SNE calculation, distance analysis,
Must enable unbiased data analysis including k-means clustering, UMAP and t-SNE calculation
Must enable distance analysis and population-distance analysis
Delivery and installation must be included
Basic Operational Training for 2 days on site with at least 2 participants
Advanced analytical and experimental design Training for 3 days on site with at least 2 participants
Warranty incl. full service / maintenance for 12 months is included
-. a quotation (optional) for a full-service maintenance contract (bronze and silver (or equivalent)) following the warranty period with a term of four (4) years.

Key features and unique system characteristics:
1.Ultrahigh-content analysis of hundreds of markers persample on multiple samples at a time

6.Sequential staining with 3 antibodies per staining cycle ensures that there is no steric hindrance of binding clones (versus binding all antibodies at once in the first step as seen on other platforms)
7.The system offers two ways of removing fluorescent signals: photobleaching by a powerful light source and enzymatic treatment is possible and can be used independently and in combination.
8.Extensive catalog of >2000 prevalidated antibodies in ready-to-use formats, tested on different samples such as formalin-fixed, paraffin-embedded (FFPE) tissue sections as well as fresh-frozen tissue sections fixed with either paraformaldehyde (PFA), or acetone:
oThe availability of a larger set of manufacturer validated antibodies and panels reduces the time for reagent optimization and thus helps to speed up discovery.
9.Open-source reagent supply: directly conjugated antibodies (FITC/PE/APC) raised in any species and from any vendor on the market can be used to build own panels or add as drop-in to existing panels provided by Miltenyi. This allows easy panel building and opens the entire available pre-labelled antibody market to a user of this platform.
10.Protein-Expression analysis at subcellular resolution (<600nm) for analysis at single cell level
11.Specially developed MACSwell Sample Carriers and MACSima Buffers to process any kind of fixed sample, including cell suspension and tissue sections (Patent EP3711859A1_MACSwell) Imaging area of the MACSima is max. 17 x 47,25 mm allowing measurement of larger tissue sections with different morphologically relevant cell types in one run, as well as complete TMAs. Furthermore the MACSWell samples are designed in a way that allow removal of the slide mounted samples without damaging the tissue. (Patent EP3711859A1_MACSwell)
12.Four different carriers can be mounted and run simultaneously for faster delivery of complete data sets in a single run
13.CAR Detection Reagents allow the detection of CAR-T cells in histological sections allowing direct ID and enumeration of these therapeutic cells (Patent EP3712617A1_CAR detection)
14.No instrument upgrade will be needed to transform the Miltenyi MACSima from a high-content protein detection system to a true MultiOMICs platform: A software upgrade as part of the package will allow to use the platform to detect a combination of protein and RNA (Patent WO2020207963A1_REAL_Barcode_Abs)
15.MACS iQ View Analysis Software offering powerful tools for complex data analysis, interactive and intuitive user interface, and easy data extraction to generate multiple graphs and plots:

VI.4.3) Application of legal remedies Detailed procedures on deadlines for the application of legal remedies: According to § 160 (3) Clause 1 German Act against Restraints of Competition (GWB) an application for review is inadmissible if 1. the applicant became aware of the claimed violation of public procurement provisions before filing the application for review, but did not complain to the contracting authority within a time limit of 10 calendar days; the expiry of the time limit under § 134(2) remains unaffected, 2. violations of public procurement provisions which become apparent from the tender notice are not notified to the contracting authority by the end of the time limit for the application or the submission of a tender specified in the notice 3. violations of public procurement provisions which only become apparent from the procurement documents are not notified to the contracting authority by the end of the time limit for the application or the submission of a tender specified in the notice, 4. more than 15 calendar days have expired since receipt of notification from the contracting authority that it is unwilling to redress the objection. Sentence 1 shall not apply to an application under § 135(1) no. 2 to have the contract declared ineffective. § 134(1) sentence 2 shall remain unaffected. According to § 134 (1) German Act against Restraints of Competition (GWB) public contracting authorities shall inform the unsuccessful tenderers in writing and without delay of the name of the successful undertaking, the reasons for the rejection of their tenders and of the earliest date of the conclusion of the contract. This shall also apply to candidates who were not informed of the rejection of their tenders before notification of the decision on the award was sent to the successful tenderers. According to § 134 (2) German Act against Restraints of Competition (GWB) a contract may be concluded at the earliest 10 calendar days after the information pursuant to paragraph 1 has been sent (by fax, e-mail, electronically or via the e-tendering online portal. The standstill period shall begin on the day after which the contracting authority dispatches the information; the date of receipt by the tenderer and candidate in question shall be irrelevant.
